     This code shows how to create a multimedia type rollover menu that changes text and graphics on the form. This uses a rect function to recognize the rollover hotspots. (A MUST SEE)

Looks good.

The proper way is to use
Set/ReleaseCapture functions to select/deselect buttons though.

Why does it have the same thing done twice, using the API and the pure VB code?
The Function CursorOnButton in the Form_MouseMove sub does the same that the PtInRegion in imgButton_MouseDown.
I think that either delete the regions and the related API functions using:
If intButton = 0 Then
and pass the initButton declaration to the form level instead of
If PtInRegion(hRectRgn(0), X + imgButton.Left, Y + imgButton.Top) Then
in imgButton_MouseDown sub for every button.
Or, if you want to stand with API the CursorOnButton function can be changed to use the PtInRegion API fuction.
